Med20em1(IMPC)J/Med20em1(IMPC)J mice exhibit embryonic lethality, with embryos recovered at E3.5 but not at E7.5. Blastocysts grown in vitro fail to hatch from the zona pellucida.

embryo
• in vitro outgrowth assays show that all blastocysts fail to hatch or grow normally; 2 of 9 arrest during the process of hatching
• blastocysts fail to hatch from the zona pellucida with apparent cell death after 3 days in culture (J:279207)
• 3 of 9 blastocysts fail to hatch out of the zona pellucida in outgrowth assays (J:286962)
• no epiblast cells are observed after 3 days in culture
• no primitive endoderm (PrE) cells are observed after 3 days in culture
• 5 of 7 blastocysts flushed at E3.5 and cultured overnight (71.4%) show widespread ectopic expression of NANOG (an epiblast marker) in the outer putative CDX2+ trophectoderm cells, indicating a failure to establish the trophectoderm lineage
• however, no Trp53-dependent apoptosis is observed and localization of POU5F1/OCT4 (ICM marker), CDX2 (trophectoderm marker), and SOX17 (primitive endoderm) is normal, indicating normal ICM, trophectoderm and primitive endoderm specification
• no trophectoderm (TE) is observed after 3 days in culture
• in vitro outgrowth assays show that 4 of 9 hatch free of the zona pellucida but do not form a monolayer of attached trophoblast cells
• when blastocysts are flushed at E3.5 and cultured overnight, epiblast marker NANOG is ectopically expressed in the trophectoderm, indicating defects in trophoblast specification

mortality/aging
• normal blastocysts are present in expected Mendelian ratios at E3.5; however, no embryos or excess resorption sites are observed at E7.5 (J:279207)
• homozygous embryos cannot be recovered in vivo after E3.5 (J:286962)

reproductive system
• in vitro outgrowth assays show that blastocysts fail to hatch with apparent cell death and failure to form a typical outgrowth colony after 3 days in culture (J:279207)
• no excessive empty decidua are noted at E7.5; no embryos are found during gastrulation, suggesting failure of implantation (J:286962)
