Age-related amyloid beta plaque progression in Tg(tetO/Prnp-APP*Swe*Lon)9191Krz/0 Tg(Camk2a-tTA)1Mmay/0 mice

growth/size/body
• lower body weight in old mice

homeostasis/metabolism
• mice exhibit accumulation of amyloid beta plaques with age, first seen in the cerebral cortex at 8 months of age and then in the hippocampus between 10-12.5 months of age
• plaques include both dense-core and diffuse types and comparable plaque loads are seen in the cortex and hippocampus
• mice show an age-dependent increase in the production of amyloid beta 38, 40 and 42 in the brain, with more than 90% of amyloid beta 40 building the dense cores of plaques, and amyloid beta 42 being the main component of diffuse plaques and loosely core-associated oligomers

nervous system
• mutants exhibit reduced forebrain weight compared to non-transgenic mice but similar in weight to single Tg(Camk2a-tTA)Mmay expressing mice
• mutants exhibit decreased dentate gyrus size compared to non-transgenic mice but size is similar to that seen in single Tg(Camk2a-tTA)Mmay expressing mice
• gliosis is associated with dense-core plaques
• mice exhibit aggravated axonal curvature and swollen, dystrophic neurites surrounding plaques
