• at E13.5, mice show altered differentiation of progenitor cells in the spinal cord central canal area
• embryos exhibit abnormal dorso-ventral patterning and reduced size of the neural tube
• in culture, secondary neurospheres derived from P0 spinal cords show altered distribution of ependymal proliferation markers, with Cyclin D2 and Ki67 expression observed throughout the neurosphere area instead of the outer circumference as in wild-type spheres
• at E13.5, immunostaining revealed reduced expression of nestin (neural stem/progenitor cell marker) and GFAP (neural stem cell marker) in the spinal cord
• at E13.5, Isl1/2-positive motor neuron cells are significantly decreased in the spinal cord
• at E13.5, immunostaining revealed reduced expression of nestin (neural stem/progenitor cell marker) in the central canal of the spinal cord
• at E12.5, expression of Pax6 (neural progenitor cell marker) is reduced in the central region (p0-p2) of the spinal cord
• at E13.5, the total number of Isl1/2-positive cells (motoneuron and interneuron markers) are significantly reduced in the spinal cord
• at E13.5, mice show decreased cell proliferation and altered cell differentiation in the spinal cord central canal area
• however, blood vessel formation around the central canal is normal
• at E16.5, the width and length of the spinal cord is significantly reduced at the thoracic level
• BrdU-positive cells are significantly reduced at E13.5 and E16.5, indicating decreased cell proliferation
• after 5 days in culture, the diameter, but not number, of neurospheres derived from the P6 spinal cord thoracic region is significantly reduced relative to that in wild-type spheres
