Polr1dem1(IMPC)J/Polr1dem1(IMPC)J mice exhibit embryonic lethality, with embryos recovered at E3.5 but not at E7.5. Embryos at E3.5 appear as compacted morula. Morulas fail to hatch from the zona pellucida with apparent cell death and never reach blastocyst stage in culture.

• at E3.0, all morula embryos exhibit many TUNEL+ blastomeres, indicating increased DNA damage and cell death
• however, no obvious TRP53+ cells are detected at E3.0, suggesting that cell death is not TRP53dependent

embryo
• at E3.0, all morula embryos exhibit many TUNEL+ blastomeres, indicating increased DNA damage and cell death

mortality/aging
• homozygous embryos appearing as compacted morulas are recovered in expected Mendelian ratios at E3.5 but arrest/die prior to blastocyst formation; no embryos or excess resorption sites are observed at E7.5

reproductive system
• no embryos or empty decidua are found at E7.5, suggesting failure of embryo implantation
