• at E6.5, embryos show a complete loss of Oct4, Nanog and Sox2 mRNA as well as protein, indicating loss of critical pluripotency factors
• embryos exhibit disorganization by E7.5
• embryos successfully implant and form egg-cylinder stages but fail to initiate gastrulation; no features of gastrulating embryos are noted at E6.5 or E7.5
• expression of brachyury (T) is almost indetectable at E6.5, indicating a deficiency in gastrulation initiation
• embryos reach egg-cylinder stage but do not initiate gastrulation
• embryos are significantly delayed at E6.5 and E7.5
• embryos are significantly smaller than controls at E6.5 and E7.5
• at E6.5, expression of OCT4 and NANOG (epiblast markers) is completely absent while expression of CDX2 (a marker of extraembryonic ectoderm) is normal, indicating loss of epiblast pluripotency in pre-streak stage embryos
