• qPCR analysis revealed a significant downregulation of expression of Th17-related proinflammatory cytokine and chemokine genes (Il6, Il1a, Il1b, Il17c, Il17f, Cxcl2, and S100a9) and a trend towards reduced Il17a gene expression in IMQ-treated mouse ear skin
• mice treated with imiquimod cream (IMQ, a ligand for TLR7/8) develop less severe dermatitis with significantly reduced ear lobe swelling relative to IMQ-treated wild-type controls under both cohoused and separately housed conditions
• antibiotic-treated mice show significantly less ear swelling than antibiotic-treated wild-type controls, suggesting that suppression of IMQ-induced dermatitis is not mediated by a change in commensal microbiota
• IMQ-treated mice show attenuated psoriasiform skin pathology with significantly lower redness, scaling and cumulative scores, reduced epidermal thickness, and milder hyperkeratosis, parakeratosis, spongiosis, acanthosis and microabscess formation in the ear lobes than IMQ-treated wild-type controls under both cohoused and separately housed conditions
• no significant difference in IMQ-induced dermatitis severity is noted between separate and cohoused groups
