mortality/aging
• only a few homozygotes survive to adulthood
• although homozygotes are born alive, ~33% of newborn mice die before weaning from unknown causes

reproductive system
• natural mating of homozygous pairs of mice results in a significantly smaller litter size than that obtained from mating of heterozygous pairs

cellular
• conversion of 5-methylcytosine (5mC) to 5-hydroxymethylcytosine (5hmC) is impaired in maternal zygotes: the 5mC level remains high while the 5hmC level remains low in the male pronucleus; however, injection of Mettl23 mRNA into oocytes restores the 5mC to 5hmC conversion in maternal zygotes
• immunofluorescence analysis of Tet3 (a 5mC-oxidizing enzyme responsible for active DNA demethylation) shows a significantly reduced Tet3 signal in maternal zygotes; however, injection of Mettl23 mRNA into maternal zygotes restores the accumulation of Tet3 into the male pronucleus
• only faint signals for STPG4 (sperm-tail PG-rich repeat containing 4; aka GSE) are detected in the pronuclei of maternal zygotes; however, injection of Mettl23 mRNA results in recruitment of STPG4 into the male pronucleus, indicating that maternal Mettl23 is likely responsible for 5mC oxidation by recruiting the STPG4-Tet3 complex into the chromatin during zygote development
