• ventricular cardiomyocytes show increased L-type calcium channel current (ICa,L) density relative to wild-type cardiomyocytes
• midpoint of steady-state activation (V ) is significantly shifted +4 mV relative to wild type myocytes; beta-adrenergic stimulation normalizes the shift of activation V such that isoproterenol-modulated channel voltage-dependence is no longer significantly different from wild-type controls
• despite increased ICa,L density, cardiomyocyte Ca2+ twitch transient amplitude is significantly less than that in wild-type myocytes
• however, current kinetics, steady-state inactivation and facilitation remain normal and cell shortening is not significantly altered
