• all mice die within 24 h after birth with no apparent defects in major organs

behavior/neurological
• no milk is visible in the stomach of dead pus

craniofacial
• TUNEL assays showed a significant decrease in the number of apoptotic cells in the rostral and caudal regions of the medial edge epithelium (MEE) of the palatal shelves at E14.5 and E15.5
• immunohistochemistry showed a significant reduction of SMAD2/3 phosphorylation in the rostral and caudal regions of the MEE at E14.5, indicating a defect in the TGFbeta signaling pathway
• although formation and elevation of the palatal shelves is observed in the rostral region, no midline epithelial seam is seen in the caudal region at E14.5 and palatal fusion is incomplete from E15.5 to E16.5
• a significant reduction of apoptosis is detected at the MEE seam at E14.5
• however, palatal edge epithelial cell proliferation is normal
• all mice show a cleft palate phenotype
