nervous system
N
• adult male mice show normal gross brain morphology, as indicated by NeuN immunohistochemistry of coronal sections
• after two-photon glutamate uncaging mediated single synapse stimulation, CA1 pyramidal neurons from organotypic hippocampal slices transfected with EGFP show a normal spine volume change in the transient phase but a significantly higher spine volume change in the sustained phase of spine enlargement, indicating enhanced dendritic spine structural plasticity in the hippocampus
• adult male mice (aged 4-14 months) show a significant, age-dependent increase in dendritic spine density in hippocampal CA1 neurons relative to control mice
• adult male mice (aged 4-14 months) show a significant, age-dependent increase in average apical dendritic spine density in CA1 pyramidal neurons relative to control mice
• however, spine synapse density and ultrastructure are normal in the stratum radiatum of the CA1 region, with no significant alterations in postsynaptic density (PSD) area and length, active zone length, dendritic spine head size or number of docked synaptic vesicles
• after high-frequency stimulation (3 trains of 100 Hz, separated by intervals of 20 s), long term potentiation of field excitatory postsynaptic potentials (fEPSPs) at Schaffer collateral/CA1 synapses is enhanced

behavior/neurological
N
• adult male mice show no significant differences in motor activity, coordination and balance, anxiety-like behavior, working memory, or performance in the Morris Water Maze task relative to control mice
• in the object-in-place (OIP) spatial memory task, adult male mice show significant discrimination of the familiar and novel object locations and a stronger preference for the object in the novel location than control mice, indicating enhanced hippocampus-dependent spatial memory
• however, no differences are observed in total object exploration time during OIP training

homeostasis/metabolism
• in an Arf6 activation assay, hippocampal lysates show a 300% increase in the active form of Arf6 (GTP bound) relative to control lysates, as measured by active Arf6 pulled down with the GGA3-PBD
