cardiovascular system
• although mice form a vascular plexus in the retina at P5, vascular progression toward the periphery is delayed
• vessel maturation (measured by the association of blood vessels with mural cells) is compromised, with mice showing reduced length of alphaSMA-positive vessels in P5 retinas
• the density and number of vascular branch points is increased in P5 retinas and an increase in vascular branching is seen in P15 retinas
• at the vascular front, the number of filopodia is increased in P5 retinas
• however, the distribution of anti-NG2-positive pericytes is unaffected in P5 retinas
|
• mice show defective retinal angiogenesis
|
• retina shows diffuse fibrinogen staining outside blood vessels and prominent staining on vessel walls, indicating leakage of plasma protein and perfused sulfo-NHS-LC-biotin is occasionally detected in extravascular spaces indicating reduced vessel integrity
• however, mice show intact coverage of retinal arteries with vascular smooth muscle cells and normal pericyte investment at P15
