mortality/aging
• survival is 80% at 100 days of age and 60% at 200 days of age
• adult mice with tumors placed on dox survive through 200 days
|
neoplasm
• a few mutants show disseminated tumor in the lumbrosacral region, partially encasing nerve roots in the subarachnoid spaces adjacent to the spinal cord, indicating spinal metastases
|
• spinal metastases are discontinuous with primary tumors, suggesting leptomeningeal spread through cerebrospinal fluid
|
• mice develop cerebellar tumors that show histology of classic medulloblastoma and large cell anaplastic medulloblastoma
• classic tumors consist of cells with moderate stromal content, low to intermediate vascularity, and variable anaplasia
• large cell anaplastic tumors have larger cells which show high nuclear-to-cytoplasmic ratios, polygonal cell morphology, and high levels or anaplasia and invasion which disrupts the molecular layer, Purkinje layer, and internal granular layer
• mice placed on doxycycline (dox) at P1 for 30 days do not develop tumors at P200
• acute treatment with dox results in a decrease in apoptotic cells and in cell proliferation in tumors
