Embryonic lungs from wild type (WT, A) and mutant (B) 17.5 dpc embryos were fixed, paraffin-embedded, sectioned and then immunohistochemically stained with Pecam-1 antibody followed by biotinylated secondary antibody, avidin-alkaline phosphatase complex, and BCIP/NBT substrate. Lung sections were counterstained with nuclear fast red. C-D: Foxm1 deficiency does not cause an increase in apoptosis. To detect apoptotic cells, paraffin lung sections were prepared from wild type and mutant 17.5 dpc embryos and used for TUNEL assay. E-F: Mutant lungs displayed a reduced number of pulmonary blood vessels. Hematoxylin and eosin and Pecam-1 stained lung sections from 15.5 and 17.5 dpc mutant and wild type embryos were used to count the number of pulmonary vessels in x400 microscope field. Five random lung sections were counted in four different mouse lungs to calculate the mean number of large vessels +/- S.D. A p value less than or equal to 0.05 is shown with an asterisk. G: Mutant lungs exhibit diminished Flk1 expression.
