Caption | A-D: Hematoxylin and eosin (H&E) staining of 15.5 dpc mouse lungs. Mouse lungs were dissected from wild type (WT, A and C) and Foxm1tm1.1Rhc/Foxm1tm1.1Rhc (Foxm1-/-, B and D) 15.5 dpc embryos and then were fixed and paraffin-embedded. Paraffin sections were stained with H&E. Mutant embryos displayed diminished numbers of mesenchymal cells surrounding distal pulmonary epithelium (ep, D). E-F: Mutant lungs exhibited defects in mesenchyme proliferation. Paraffin sections from wild type (E) and mutant (F) 15.5 dpc embryos were immunohistochemically stained with BrdUrd antibody followed by anti-mouse antibody conjugated with alkaline phosphatase and BCIP/NBT substrate. Lung sections were counterstained with nuclear fast red. Although BrdUrd was detected in both epithelial and mesenchymal cells of wild type lungs (E), mutant lungs exhibited only epitelial BrdUrd staining (F, shown with arrows). G-J: Immunostaining of lung sections with antibodies specific to Foxa2 and vimentin. Lung 15.5 dpc sections were stained for Foxa2 (G and H) and vimentin protein (I and J).
