Caption | A: Light microscopy. One-um sections of lung parenchyma were cut and stained with toluidine blue. Note that in Cav1tm1Mls/Cav1tm1Mls (KO) mice, the alveolar spaces appeared significantly smaller or appeared constricted, with thickened alveolar septa and hypercellularity. These images were acquired with a 60x objective. Upper panel, wild-type (WT) mice; lower panel. mutant mice. B: Reticulin staining. Wild-type and mutant mouse lung tissue sections were subjected to reticulin staining and examined using a Zeis Axiophot with a 20X objective. Note that in mutant mice, reticulin staining showed increased basement membranes in the thickened alveolar walls. There was increased density and thickness of the basement membranes and loose arrays of reticulin fibers. Upper panel, wild-type; lower panel, mutant mice. C: Transmission electron microscopy. Lung tissue samples were processed for electron microscopy. Images were acquired at low magnification (x2,000), and montages were assembled to illustrate the detailed morphology of the alveolar architecture. Wild-type and mutant mouse lung tissue sections stained with hematoxylin and eosin were examined using a Zeis Axiophot. Using a 20x objective, six random 0.5mm2 fields were photographed for each genotype, and all the nuclei within those regions were manually tabulated using a hand-held counter. Note that lung sections from mutant mice show an ~2-fold increase in overall cellularity. | ||||
