Caption | Transmission electron micrographs of embryonic day 18 kidneys from wild-type and Podxltm1Kmn/Podxltm1Kmn embryos. A: Wild-type (+/+) kidneys have normal podocytes (Pod.) with typical major processes (MPs) and foot processes (FPs) that embrace the outer aspect of the capillary basement membrane. Red blood cells (RBCs) can be seen in the lumen of the endothelial vessels. Podxltm1Kmn/Podxltm1Kmn (-/-) mice show a complete loss of slit diaphragms (SDs) and FPs. Junctional complexes (JCs) are present between all of the mutant podocytes. Podocytes in mutant mice also display numerous vacuoles (Vac.) and overall the glomerular capillaries had a thicker endothelial cell (EC) layer. Scale bars, 2 um. B: Higher magnification reveals that the SDs and FPs indicated by arrows in the wild-type and Podxltm1Kmn/Podxl+ (+/-) mice are completely lost in homozygous mutant glomeruli. JCs including tight junctions (TJs) and adherens junctions (AJs) are present between homozygous mutant podocytes.
