All images are from paraffin sections, except for h and i, which are sections of methacrylate (Technovit 7100)-embedded material. The skin biopsies were taken from female olt heterozygotes (+/olt) or olt/olt mice on postnatal day P13 (c,d), P15 (a,b,e,f,g,j,k) and from 3 month old mice (h,i). Images j and k are higher magnifications of the boxed areas in b and g. PC, panniculus carnosus; Epi, epidermis; abd muscles, abdominal muscles; MamGI, mammary gland. Hair shafts of homozygous mutants are twisted at the level of the dermis and epidermis, which involves almost all hair follicles in the lower ventral (left arrow in f) and medial hindlimb regions (d, boxed area in g, arrowed in k), but involves only a minority in the dorsal region (boxed area in b, arrows in j). In the medial femoral region, the shape of the hair bulbs indicates transition from anagen on P13 (arrowed in d; compare with control, arrows in c) to catagen on P15 (arrows in g; compare with control, arrowed in e). The majority of hair follicles in the dorsal skin show anagen stage of development on P15 in the mutant (arrowed in b; compare with control, arrows in a). After 3 months of age, hair follicles are lost from the mutant medial hind limb, with some hair bulb remnants (small arrow in i) in the subcutis next to normal-appearing telogen follicles (long arrow in i), while in the control, all hair follicles are synchronously in telogen (arrow in h). | ||||
