A, gross phenotypes of Pkd2tm1Gwu/Pkd2tm1Gwu (Pkdnf3/nf3) and Pkhd1tm1Gwu/Pkhd1tm1Gwu (Pkhd1-/-) mice demonstrated growth retardation compared with wild-type (WT) littermates 2 weeks after birth (panel a). Severe dilatation of the common bile duct (arrow) was observed in Pkd2tm1Gwu/Pkd2tm1Gwu mice (panel b). The gross cystic phenotypes (arrows) were similar between 12 month old Pkd2tm1Gwu/Pkd2tm1Gwu kidneys (panel c) and 4 month old Pkhd1tm1Gwu/Pkhd1tm1Gwu kidneys (panel d). In the same pair of mice, similar gross cystic phenotypes and focal necrosis (arrows) were seen in the liver (panels e and f). B, microscopic phenotypes of Pkd2tm1Gwu/Pkd2tm1Gwu and Pkhd1tm1Gwu/Pkhd1tm1Gwu mice. In 12 month old wild-type mice, medullary tubules (panel a), cortical glomeruli (panel b), and the tubulointerstitial compartment (panel b) were normal. Similarly, liver (panel c) and pancreatic (panel d) sections exhibited normal histology in the same mouse. In 12 month old Pkd2tm1Gwu/Pkd2tm1Gwu mice, there was a focal dilatation of papillary collecting ducts and patchy dilatation of proximal tubules (panels e and f), as well as an increased expansion of Bowman's space (right arrow in panel f) in the renal cortex. These mice also showed dilated biliary ducts and central veins with lymphocyte infiltration/necrosis (panel g, arrows). There was also dilatation of both small and large pancreatic ducts (panel h, arrow). Tissue sections from 4 month old Pkhd1tm1Gwu/Pkhd1tm1Gwu mice, which correspond to panels e-h, are shown in panels i-l. Scale bars = 2 mm in A and 10 um in B. | ||||||
