Caption | Histology of seminiferous tubules. Histological analyses of testicular sections of wild-type (+/+), Hsf1tm1Miv/Hsf1tm1Miv (hsf1-/-), Hsf2tm1Miv/Hsf2tm1Miv (hsf2-/-), or Hsf1tm1Miv/Hsf1tm1Miv Hsf2tm1Miv/Hsf2tm1Miv (hsf1-/- hsf2-/-) adult (12 weeks) mice. A: Paraffin-embedded sections were stained with H&E. Testes from Hsf2tm1Miv/Hsf2tm1Miv mice show seminiferous tubules with disruption of spermatogenesis (marked by arrow) such as reduction of spermatocytes and spermatids. Note the severe testicular tissue degeneration such as reduced number of spermatogonia, complete absence of developing spermatocytes and spermatids, tubular vacuolization, and formation of multinuclear giant cells (right panel, marked by arrow) in testes of double homozygous mice. Note the Sertoli cells visible at the basal layer in testes of double homozygous mice (right panel); no visible abnormalities were observed in testes of Hsf1tm1Miv/Hsf1tm1Miv or wild-type mice. Testicular sections were viewed at relatively low x200 (left panels) or high x630 (right panels) magnification. B: Epididymal sections show a reduction (Hsf2tm1Miv/Hsf2tm1Miv) or complete absence (double homozygotes) in mature spermatozoa. In contrast, mature spermatozoa are abundantly present in epididymis of Hsf1tm1Miv/Hsf1tm1Miv or wild-type mice. x630. C: In situ detection of apoptotic cells in sections from seminiferous tubules from wild-type, Hsf1 homozygous mutant, Hsf2 homozygous mutant and double mutant mice by TUNEL assay. The size and the location within the seminiferous epithelium of the TUNEL positive cells is consistent with spermatocyte cell apoptosis. Clusters of apoptotic cells (dark red) indicated by arrows are shown on paraffin-embedded sections of testes from Hsf2 homozygotes and double mutant mice. Nuclei were counterstained with hematoxylin. x400. | ||||||
