Caption Hemophagocytes in bone marrow of Lgmntm1Ihn/Lgmntm1Ihn mice increases with age. (A-C) H&E staining of bone marrow from Lgmntm1Ihn/Lgmn+ (+/-) and Lgmntm1Ihn/Lgmntm1Ihn (-/-) mice at ages of 3 months and 12 months (A). Bone marrows from 12 month old homozygous mutant mice were further analyzed under high magnification, and many erythrocytes were engulfed in histiocytes (B), and morphologically Gaucher-like cells with a wrinkled appearance were observed (C). (D) The Gaucher-like cells (arrowheads) as well as hemophagocytes (arrow) in bone marrow of homozygous mutant mice showed strong autofluorescence. (E) PAS staining of bone marrow from homozygous mutant mouse. Gaucher-like cells were strongly stained. (F) Bone marrow cells of homozygous mutant mice and age matched control heterozygous mice were cytospun and stained with Giemsa, F4/80 mAb, and CD68 mAb. Hemophagocytes of mutant mice were characterized as both F4/80-positive and CD68-positive histiocytes.
