Caption (D) Schematic showing a portion of the Panx2 (Px2) genomic loci containing exon 1 (box, protein coding region in black) and the segment (bold line) that was manipulated to generate the targeting constructs. The structure of the locus for the wild-type (WT) and knockout (KO) alleles, the latter with the Neo cassette and after its excision via Cre-mediated recombination to generate the Panx2tm1.1Mony allele, is illustrated. Also shown are the relevant restriction endonuclease sites, the locations of the LacZ and loxP-flanked neomycin cassettes within the targeting constructs and knockout alleles, the locations of DNA fragments used to generate probes for Southern blot analysis, and the origins of the fragments that helped in determining the genotypes of mice and embryonic stem cells.
