Caption Trophoblast giant cells (TGCs) in wild-type (WT Fem) and heterozygous Plac1tm1(KOMP)Vlcg/Plac1+ females that inherit the mutant allele from the mother (Xm-X) and in homozygous Plac1tm1(KOMP)Vlcg/Plac1tm1(KOMP)Vlcg females that inherit one mutant allele from the mother and the other mutant allele from the father (Xm-Xp-). In situ hybridization of Prl2c2 expression using the digoxygenin detection system. Increased TGCs numbers were evident in the labyrinth layer of females inheriting the mutant allele from the mother. Black arrows = partial TGCs adjacent to the decidua; Green arrow = sinusoidal TGCs; Red arrow = spongiotrophoblasts at the interface of the junctional zone and labyrinth. Non-specific signal was determined by hybridization with the "sense" riboprobe construct. Magnification = 1.5x.
