Caption | PikfyveGt(AT0926)Wtsi/PikfyveGt(AT0926)Wtsi (PikfyveBeta-geo/Beta-geo) mutants that survive past 2 weeks are smaller than their heterozygous littermates and develop extensive neurodegeneration. (A) Homozygous P16 pups were half the weight of their heterozygous (het) littermates. Seven heterozygotes and homozygotes were analyzed. (B) At P16, the brains of the homozygous mice had a normal architecture but were smaller than those of their heterozygous littermates. (C) The brains of the homozygous mutant mice weighed ~30% less than those of their heterozygous littermates. Two heterozygotes and homozygotes were analyzed. (D) Sagittal sections of the brain and coronal sections of the cerebellum of P16-P19 mice stained with H&E revealed prominent vacuolation in the homozygous brainstem, notably the midbrain and pons. The deep cerebellar nuclei are affected also. (E) The spinal cords of the P19 homozygous mutants stained with H&E revealed the presence of large vacuoles. | ||||
