Palatal shelves of Map3k7^tm1Mis/Map3k7^tm1Mis H2az2^{Tg(Wnt1-cre)11Rth}/0 (Tak1^FX/+/Wnt1-Cre^-) elevate and fuse in vitro, but show delayed rugal formation in vivo. A and B, control (left panels) andmutant (right panels) embryos were harvested at E13.5, and dissected midfacial segments (without mandible, tongue, and brain) were cultured under chemically defined conditions in roller bottles for 24 h (A) or 48 h (B). The arrows point to the tips of elevated palatal shelves in both control and mutant samples cultured for 24 h (n = 3). C and D, comparison of Shox2 and Tbx22 (C, E14.0) and Shh (D, E14.5 and E15.5) expression in control and mutant palatal regions.
