Transmission electron micrographs of tendons from 1 month-old wild-type (+/+) and mutant mice are shown. A, flexor digitorum longus tendons. Low magnification images (panels a and d; scale bar, 1 um) show disorganization of cell and matrix in the mutant tendon (panel d) compared with the wild-type tendon (panel a). At high magnification, collagen fibrils from the wild-type mice (panels b and c) display normal cross-sectional profiles in both central (panel c) and pericellular (panel b) regions. The fibrils are circular with a heterogeneous distribution of fibril diameters. In contrast, the mutant tendons (panels e and f) have larger numbers of smaller diameter fibrils (arrows) in both central (panel f) and pericellular (panel e) areas. Very large, twisted, and structurally abnormal fibrils (arrowheads and curved arrow) are found in the pericellular (panel e), but not central (panel f), areas. Scale bars, 100 nm. | ||||
