Caption | (D) Electron micrographs at three different magnifications in rhomboid muscle fibers from Fat1Gt(KST249)Byg/Fat1Gt(KST249)Byg Tg(Myl1-lacZ)1Ibdml/0 (Fat1LacZ/LacZ) adult mice at early symptomatic stages (6-15% weight loss) show fragmentation of the myofiber architecture and loss of t-tubule integrity. In wild-type (WT) myofibers, t-tubules (purple arrows) are visible between myofibrils, precisely aligned on either side of each Z-band, at a position coinciding with the end of the myosin filaments. By contrast, in dystrophic fibers from mutant mice, the general disorganization correlated with missing (stars), mis-oriented, mis-aligned (orange arrows), or fragmented (red arrows) triads. An increased distance (indicated as blue double arrowed bar) between the sarcolemma and contractile apparatus is observed in mutant muscles, compared to WT, indicating a loss of the tight association between the contractile apparatus and the sarcolemma. Scale bars: 15 um; (D1, 2) 5 um; (D3, 4) 0.5 um; (D5, 6) 0.2 um. | ||||||
