Histological analysis of 6.5-dpc embryos from Sox2^tm1.1Vep/Sox2⁺ (Sox2^betageo2/+), Sox2^tm1.1Vlcg/Sox2⁺ (Sox2^COIN/+), Sox2^tm1.2Vlcg/Sox2⁺ (Sox2^INV/+) intercrosses. Wild-type (A), Sox2^tm1.1Vep/Sox2⁺ (Sox2^betageo2/+) (B), Sox2^tm1.1Vlcg/Sox2⁺ (Sox2^COIN/+) (D), Sox2^tm1.1Vlcg/Sox2^tm1.1Vlcg (Sox2^COIN/COIN ) (E), Sox2^tm1.1Vlcg/Sox2^tm1.1Vep (Sox2^{COIN/betageo2}) (F), and Sox2^tm1.2Vlcg/Sox2⁺ (Sox2^INV/+) (G) are all phenotypically normal. In contrast, Sox2^tm1.2Vlcg/Sox2^tm1.2Vlcg (Sox2^INV/INV) (I) and Sox2^tm1.2Vlcg/Sox2^tm1.1Vep (Sox2^INV/betageo2) (H) embryos form disorganized extraembryonic tissues, but fail to form the epiblast, effectively phenocopying Sox2^tm1.1Vep/Sox2^tm1.1Vep (Sox2^{betageo2/betageo2}) embryos (C), and displaying the phenotype previously described for Sox2 homozygous-null embryos. Sections were stained with hematoxylin and eosin (H&E).
