Caption | Spon1tm1Gdpa/Spon1tm1Gdpa (KO) mice tibiae exhibit increased staining of markers of bone synthesis and turnover. Immunohistochemical staining of mid-saggital sections of tibia from 6 month old wild-type (WT) and KO mice with antibodies for F-spondin (SPON1), Alkaline Phosphatase (ALPL), Periostin (POSTN), and tartrate resistant acid phosphatase (TRAP) as a marker of osteoclast activity. Sections were counterstained with either 1% methyl green (TRAP) or hematoxylin. Arrrows indicate areas of increased staining in the epiphyseal cartilage and adjacent trabecular bone of mutant tibiae. Adjacent graphs correspond to quantification of positive stained area (SPON1, ALP, POSTN) and osteoclast number per field (TRAP). Quantification was performed on 4 sections per specimen, and values represent the average of 4 mice per genotype. * = P<0.05. Scale bars; 200 um for antibody stainings and 100 um for TRAP staining. | ||||
