Decreased focal adhesions (FA) number and size in Arhgef28^tm1.1Ddsr/Arhgef28^tm1.1Ddsr (Rgnef-/-) mouse embryo fibroblasts (MEFs) on fibronectin (FN) at 120 min. (A) Wild-type (Rgnef+/+, WT) and mutant MEFs were plated onto glass coverslips pre-coated with FN (10 mg/ml). Cells were fixed and analyzed for FA formation by paxillin staining (green), f-actin (phalloidin, red), and nuclei were stained with Hoechst (blue) as shown in a merged image. Scale is 10 um. Significantly-reduced adhesion number (B) and size (C) in mutant compared to normal (WT) MEFs. Paxillin-stained points were enumerated as FAs by thresholding images using Image J (v1.4) and FA size (pixels) was determined using Cell Profiler (v2.0). Box-and-whisker plots show distribution of the data: mean (black square), median (middle line), 25th percentile (bottom line), 75th percentile (top line), and 5th or 95th percentiles (whiskers),* p<0.05.
