In hematoxylin-eosinstaining, the morphology of megakaryocyte in Myh9tm1.1Dash/Myh9+ (R702C+/-) mice was not different from that of wild-type (C57BL/6j; WT) mice (A) (B). There were no significant differences in the distribution of NMMHCIIA between heterozygous mutant mice and WT mice by immunostaining (C) (D). Proplatelet formation was explored by cultured fetal liver cells. In WT mice, the proplatelet shaft extends from the cell spindle, leading to the formation of proplatelet beads. Heterozygous mutant mice had shorter and thicker shafts when compared with WT mice, and the proplatelet beads were fewer and larger than those of WT mice. Megakaryocytes were stained with May Grunwald Giemsa (MGG) (E) (F) or were observed using CD41 immunofluorescence (G) (H). | ||||
