Caption | Brca2tm1Brn/Brca2tm1Brn Tg(Pbsn-cre)4Prb/0 (Brca2F/F;PBCre4), Brca2tm1Brn/Brca2tm1Brn Trp53tm1Brn/Trp53+ Tg(Pbsn-cre)4Prb/0 (Brca2F/F;Trp53F/+;PBCre4), and Brca2tm1Brn/Brca2tm1Brn Trp53tm1Brn/Trp53tm1Brn Tg(Pbsn-cre)4Prb/0 (Brca2F/F;Trp53F/F;PBCre4) mutants have increased DNA damage and apoptosis. A) Immunohistochemistry for gammaH2AX shows several cells have DNA damage in low-grade prostate intraepithelial neoplasia (LG PIN) areas in Brca2tm1Brn/Brca2tm1Brn Tg(Pbsn-cre)4Prb/0 and Brca2tm1Brn/Brca2tm1Brn Trp53tm1Brn/Trp53+ Tg(Pbsn-cre)4Prb/0 prostates, whereas control (Brca2F/F) prostates have undetectable levels of DNA damage. Brca2tm1Brn/Brca2tm1Brn Trp53tm1Brn/Trp53tm1Brn Tg(Pbsn-cre)4Prb/0 mice have multi-focal areas with a large number of cells with DNA damage in regions of HG PIN. (B) TUNEL assay on sections of prostates demonstrates Brca2tm1Brn/Brca2tm1Brn Tg(Pbsn-cre)4Prb/0 and Brca2tm1Brn/Brca2tm1Brn Trp53tm1Brn/Trp53+ Tg(Pbsn-cre)4Prb/0 mutant prostates have an increased level of apoptosis, compared to control prostates. The level of apoptotic cells increases further in HG PIN lesions of Brca2tm1Brn/Brca2tm1Brn Trp53tm1Brn/Trp53tm1Brn Tg(Pbsn-cre)4Prb/0 mutants. Arrows indicate apoptotic cells. The anterior prostates of 16-month-old animals are shown. | ||||||||
