Caption | (A, C, E, G), H&E stained liver sections from Notch1tm3(cre)Rko/Notch1+ (N1::Crelo/+) animals; (B, D, F, H, I, J), H&E stained liver sections from Notch1tm3(cre)Rko/Notch1tm2Rko (N1::Crelo/fl) animals. Even at low magnification, clear differences are apparent between mutant (B) and control (A) mice. (C,D) Close up of the portal triads, consisting of the portal vein, the bile duct and an artery. In mutant liver, dense chronic inflammatory infiltrates are present around the bile ducts (black arrows in B and D); in addition, the hepatic cords are more separated, and there appears to be some modest hypercellularity between the hepatic cords in multiple areas (B). At higher magnification, dense chronic inflammatory infiltrates around the bile duct are evident in mutant liver (D, arrow). (F) shows the hepatic cords. Note that in many areas in the mutant liver, the hepatic cords are separated as the sinusoids are expanded by a dense chronic inflammatory infiltrate (F, arrow). (G, H) Blood enters the liver at the portal triad, flows through the sinusoids between the hepatic cords and drains finally into the central vein (shown here). The central vein in the mutant liver has modest expansion and the cellularity within the sinusoids separating the hepatic cords is also increased (H) compared to that of the control liver (G). The mutant livers also have areas with blood-filled cavities (I), some of which contains big thrombi (J). Scale bars represent 50 um. | ||||||
