Caption Sprouting of secondary neurites but not the formation of neuromuscular junctions is affected in Srrm4tm1b(EUCOMM)Wtsi/Srrm4tm1b(EUCOMM)Wtsi (delta7-8/delta7-8) mice. A. Whole-mount immunofluorescence of E16.5 diaphragms from heterozygous and homozygous mutant embryos using an antibody to neurofilament (green) highlights defective innervation in homozygous mutants. B. The length covered by secondary branches and (C) their numbers are already defective in homozygous mutants at E16.5. Three diaphragms were analyzed for each genotype. D. The total length covered by primary branches is not affected at E16.5. E. The average length of individual secondary branches in the mutants is the same as wild-type and heterozygous littermates at E16.5. Three diaphragms were analyzed for each genotype. F. Whole-mount staining of E18.5 diaphragms from heterozygous and homozygous mutant mice using an anti-neurofilament antibody (green) and Alexa-594-labelled alpha-bungarotoxin (red) to highlight innervation and motor endplates. Lower panels show higher magnification of boxed regions. Scale bars: 1000 um in upper panel, 100 um in inset. G. The number of endplates observed on a 475 um-long segment was quantified on the left ventrally-projecting nerve of the diaphragm and (H) the distance between endplates and the primary branch was measured (right). Four heterozygous or five homozygous diaphragms were analyzed per genotype. Whiskers indicate 10th and 90th percentiles in all box plots. One-tailed Mann-Whitney test.
