High magnification fluorescent images of the pharyngeal arch region of E9.5-10.5 DAPI stained (a) wild-type and (b) pac/pac (pacman) embryos. Arrowheads illustrate the abnormal size and shape of individual pharyngeal arches in each of the mutants identified in our screen. Mutant embryos at E9.5 display rostral and caudal pharyngeal arches that are distinct from each other but exhibit abnormally expanded mesoderm derived arch arteries. The expanded arch artery is particularly evident within the first pharyngeal arch giving it the appearance of being massively enlarged and also hollow.
