Caption Bright field images of Sox10 in situ hybridization stained E9.5-10.5 (a) wild-type and (b) Aldh1a2grim/Aldh1a2grim (grimace) embryos. Arrowheads and arrows, respectively point to ectopic Sox10 positive ganglia and agenesis or hypoplasia of the neural crest cell component of the cranial ganglia. Mutant embryos exhibit severe defects in cranial ganglia development. The trigeminal ganglion is hypoplastic. The spatiotemporal reduction in Sox10 staining more caudally is consistent with agenesis of the facioacoustic/vestibulochoclear, glossopharyngeal, and vagal ganglia. The circular domains of Sox10 staining that remain coincide with small ectopic supernumerary otocysts.
