Caption Bright and fluorescent images of Neurofilament of beta-tubulin innunostained E9.5-10.5 (a) wild-type and (b) Aldh1a2grim/Aldh1a2grim (grimace) embryos. Arrowheads and arrows highlight agenesis or hypoplasia of neuronal differentiation in the midbrain and cranial ganglia. Neurofilament staining reveals near complete nervous system agenesis. Except for a small amount of neurofilament staining at the isthmus, there is no evidence of the neuronal network or plexus that typically develops in the midbrain (arrowhead). Defects in peripheral nervous system development are similarly extreme. Except for a couple of very discrete localized domains of neurofilament staining adjacent to the hindbrain, there is little evidence of any overt neural differentiation in association with the cranial ganglia. Abbreviations: pl, plexus; f, facioacoustic/vestibule cochlear; g, glossopharyngeal; tg, trigeminal; v. vagal.
