Analysis of hypertrophic zone width in wild-type (wt), Col10a1^tm1.1Rpbh/Col10a1⁺ (wt/m) and Col10a1^tm1.1Rpbh/Col10a1^tm1.1Rpbh (m/m) mice. (A) H & E staining of tibial growth plates from new born, 3 week and 7 week old mice. The hypertrophic zone is indicated by the vertical red brackets; horizontal white scale bar = 100 um. The hypertrophic zones of tibial growth plates are expanded in homozygotes. In heterozygotes, a 35% expansion of the hypertrohpic zone is seen at 3 weeks but resolves by 7 weeks of age. (C) 5-bromo-29-deoxyuridine (BrdU) labelling of proliferative cells in the growth plate of 3 week old mice. Positive cells stained black. No significant differences in the chondrocyte proliferation rates is seen.
