Caption | Vestibular hair cells specifically degenerate in the saccule of Tmprss3m1Ingm/Tmprss3m1Ingm mice. Whole mount of cristae (A), utricles (B) and saccules (C-D) were immunostained for Myosin VIIa to mark hair cells (blue in upper panel, white in middle panel) (A-C), and labelled with fluorescent phalloidin to stained actin and visualize hair bundles (red in upper panel, white in down panels) (A-D). In adult cristae (A) and utricles (B) of mutant mice, normal organization and density of hair cells and hair bundles were observed. In opposite, in the saccule of mutant adult mice, very few hair cells left and almost no hair bundles were observed. (D) At P0, P10 and P27, saccular hair bundles were stained in developing mutant mice to follow and quantify a potential degeneration of hair cells during this period. No difference in hair bundle density was observed during this developmental period of the saccular epithelium between these different postnatal days. Scale bars = 100 um. | ||||
