Caption Polr1atm1b(EUCOMM)Hmgu/Polr1atm1b(EUCOMM)Hmgu embryos cannot be recovered at E7.5 (0/29), nor are there excess resorption sites. Homozygous mutants are recovered at E3.5 and appear as dying 2-cell stage embryos, suggesting zygotic genome activation failure. No further development occurs, with mutants never reaching a morphological blastocyst stage. RT-PCR indicates robust expression. Knock-in LacZ also shows robust expression in gastrulation stages.
