(A) The targeting vector inserted a FRT-flanked neo cassete with a 3' loxP site upstream of exon 2 and an additional loxP site downstream of exon 3, to generate the Ppm1a^tm1Xya allele (Targeted Allele). The targeting vector deleted a 6 kb genomic sequence containing exons 2 and 3 with the translation start codon (ATG) of Ppm1a to generate the Ppm1a^tm1.1Xya allele (Knockout Allele). The 3' external probe for Southern blot analysis was indicated. Arrows indicated positions of genotyping primers.
