Caption | Generation of the Slc6a3tm1.1Jaja hypomorphic allele. (A) Mutation of the five serines to alanine (S/A) in the N terminus of Slc6a3 were introduced into the mouse Slc6a3 gene by homologous recombination in mouse ES cells. A knock-in mouse line, Slc6a3tm1.1Jaja, was generated by injecting the positive ES cells into blastocysts of C57BL/6J mice. The targeted Slc6a3 gene had a LoxP-flanked selection marker (a neomycin resistance gene with its promoter and poly (A) signal) inserted in exon 2. To minimize potential interference with Slc6a3 expression, the selection marker was removed by crossing with mice that overexpressed Cre recombinase. PCR and Southern blotting indicated the correct homologous recombination. The modified region of the gene was PCR amplified and sequenced, confirming the presence of the intended modifications. Open blue box: mouse Slc6a3 exon 2; red line: mutation S to A at positions 2, 4, 7, 12, and 13 at the N terminus; open arrows: Loxp sites; filled box: neo cassette; open box: probe for Southern blot. | ||||
