Caption Generation of Grin1tm1Mim mice by homologous recombination in C57BL/6 strain derived ES cells. A, Schema of the exons 11-22 region of the Grin1 (GluRzeta1) gene, targeting vector, floxed and neo-inserted allele, and floxed allele (Grin1tm1Mim) allele. Exons 19 and 20 encode the putative transmembrane segment M4 of Grin1. The floxed and neo-inserted allele (GluRzeta1flox;neo allele) contains two loxP sequences flanking exons 19 and 20 of the Grin1 gene and the neo gene flanked by two frt sequences. The neo gene was removed in vivo by crossing the floxed and neo-inserted allele mice with FLP66 mice carrying the Flp recombinase gene under the control of the EF1alpha promoter. The floxed Grin1tm1Mim/Grin1+ mice were crossed with Grik4tm1(cre)Mim/Grik4+ (GluRc1-Cre) mice to disrupt the Grin1 gene selectively in the hippocampal CA3 region in J:144994 or can be crossed with any cre-expressing mice to generate conditional disrupiton of Grin1.
