In the CC10-tTs-hGH construct, the Clara Cell 10-kD protein promoter (Scgb1a1; CC10 Promoter) was used to drive the tetracycline-regulated transcriptional silencer (tTS). In the CC10-rtTA-hGH construct, the Scgb1a1 (CC10) promoter drives the reverse tetracycline-controlled transcriptional activator (rtTA). The tetO-CMV-YKL-40-beta-globin construct contains multimers of the tet operator, a minimal CMV promoter, and a human CHI3L1 (YKL-40) cDNA. All three constructs were simultaneously injected into fertilized eggs to generate leak-free triple transgenic mice. | ||||
