Generation of the Nelfb^tm1.1Roli and Nelfb^tm1.2Roli (KO) alleles. A. Illustration of the wild-type Nelfb (Cobra1) locus (top) and a portion of the targeting construct (bottom). The short (left) and long (right) homology arms encompass genomic regions 1.1 kb upstream of exon 1 and approximately 84 kb downstream of exon 4, respectively. Also indicated are loxP (red arrows), FRT sites (crescents), exons (open bars), BamH1 sites (B), promoter (solid arrow), 5' probe (blue bar), and PCR primers (block arrows). The generate the Nelfb^tm1.1Roli allele, the neo cassette was removed via flp mediated recombination, leaving exons 1-4 floxed. The floxed exons 1-4 were removed via cre-mediated recombination to generate the Nelfb^tm1.2Roli allele.
