Caption Generation of the floxed Nr0b2tm1Mjev (Floxed SHP allele) and deleted Nr0b2tm1.1Mjev (Deleted SHP allele) alleles. The top line shows a diagram of the wild-type allele with the coding exons denoted by filled boxes. Exon 1 contains the translation start site. The second line diagrams a successfully targeted conditional deletion allele in which LoxP sites (open triangles) have been inserted flanking the Nr0b2 exon 1 by the use of a neomycin resistance gene (Neor) driven by the phosphoglycerate kinase promoter (PGK). The bottom line depicts the deleted Nr0b2 allele resulting from breeding with transgenic mice expressing Cre recombinase. The positions of the primers F1, R1, and R2 used for genotyping mice are shown by arrows.
