| Caption | Generation of the Sox17tm4.1(icre/ERT2)Heli allele. (a) Targeting strategy for the allele. A targeting vector was used to introduce the viral 2A peptide followed by cre/ERT2 into exon 5 of the Sox17 gene. The Frt-flanked PGK-driven neomycin resistance (NeoR) was removed by Flp-mediated excision. Sox17 exons are numbered (1-5). 5' and 3' UTRs are shown in black, the open reading frame in orange, and predicted promoter regions and transcriptional start sites in yellow and red, respectively. Genotyping primers - EP400, EP401, EP980, and EP064. The 5' probe, restriction sites for EcoRV, 5' and 3' homology regions (retrieval) are indicated. | ||||
