Caption Generation of the conditional Nox1tm1.1Anrt allele. (A) A loxP site was inserted upstream of exon 13. An FRT-flanked neo cassette with a 3' loxP site and modified exon 13 with an in frame lacZ gene was also inserted generating the homologous recombinant in A. Flp-mediated recombination removed the neo cassette and left exon 13 floxed (homologous recombinant B) generating the Nox1tm1.1Anrt allele. Nox1 activity was abolished by truncating exon 13, the last exon of the coding sequence, eliminating FPR1-Nox1 dependent redox signaling and mucosal wound repair part of the NADPH binding site. The truncated, inactive Nox1 gene was fused to an active Beta-galactosidase. Cre-induced recombination can be used to delete the floxed exon 13 (C) and results in a beta-gal in frame, fused with inactive Nox1.
