| Caption | Generation of the Tdgtm1Psch allele. Gene Targeting of Tdg on Chromosome 10. The targeting construct was generated by replacement of exons 6 and 7 with a neomycin resistance cassette. This was achieved by the substitution of a NarI-PacI fragment with the resistance cassette in a subcloned genomic region spanning from intron 4 to the 3' UTR. In addition, the targeting construct contained a thymidine kinase (TK) cassette for negative selection. Arrowheads show the position of primers used for multiplex-PCR genotyping. The expected length of PCR products are 1.2 kb and 1.5 kb for the wild-type (wt) and the targeted loci, respectively. | ||||
