Caption Generation of Gja10tm2.1(cre)Kwi A. Part of exon 2 was replaced by the HBEGF/EGFP fusion construct flanked by frt sites (DTRfrtCre), and the NLS-Cre recombinase sequence and followed by a neomycin resistance marker (neoR). The NLS-Cre coding sequence is placed under the control of the Gja10 (Cx57) promoter after Flp recombination. DTR-eGFP: diphtheria toxin receptor (DTR; HBEGF) and EGFP fusion protein; NLS: nuclear localization signal. B. Southern blot analysis. 7.2 kb band confirms correct targeting of cre construct to the Gja10 (Cx57) locus. Successful Flp-mediated excision of DTR produces 8.7 kb band.
