Strategy for generation of Rax^{tm1.1(cre/ERT2)Sbls}. A targeting vector containing the cre recombinase coding sequence fused to a tamoxifen-inducible mutated estrogen receptor ligand binding domain (ER) sequence and Frt-flanked pGK-neo selection cassette was inserted by homologous recombination at the position 6 bases downstream of the start ATG codon of the endogenous Rax locus. Correct integration in ES cells was verified by PCR using primers P1 (R) in the neo cassette and P2 (F) in intron 2 of Rax, as indicated in the figure. The neo cassette was deleted from the germline by mating animals carrying the neo-in allele with Flp-expressing mice.
